目的:观察坐骨神经分支选择性损伤模型大鼠脊髓背角内5-羟色胺4—7受体亚型mRNAs的表达变化。 方法:实验于2004—10/2005-06在第四军医大学解剖学教研室进行。①取SD大鼠27只,随机分为正常对照组3只;手术组24只。②手术组大鼠在戍巴比妥钠(40mg/kg)腹腔麻醉下,切开左侧后肢皮肤暴露并钝性分离腓肠肌两头,暴鼹坐骨神经,在邻近坐骨神经分叉处结扎胫神经和腓总神经,于结扎远端0.5cm处切断神经,建立坐骨神经分支选择性损伤大鼠模型,全过程中注意勿伤及腓肠神经;右侧为假手术对照.只暴露不结扎切断坐骨神经,其余过程同手术侧。③分别在术后1,2,3,4,7,14,21,28d,取b~k节段脊髓背角,采用反转录-聚合酶链式反应技术检测脊髓背角内5-羟色胺4—7受体亚型mRNAs的表达变化。 结果:27只大鼠进入结果分析。①正常大鼠脊髓背角检测到5一羟色胺4、5-羟色胺5A、5-羟色胺7受体亚型mRNAs的表达,但未检测到5-羟色胺5B.5-羟色胺6受体亚型mRNAs的表达。②手术组手术侧检测到5-羟色胺4、5-羟色胺5A和5-羟色胺7受体亚型mRNAs的表达上调。其中,5-羟色胺4受体亚型mRNA的表达在术后4d明显升高,21d达高峰后下降,28d趋于正常。5-羟色胺5A受体亚型mRNA的表达在术后3d开始升高,持续升高至28d。5-羟色胺7受体亚型mRNA的表达变化同5-羟色胺5A受体亚型。假手术侧的脊髓背角,各受体亚型mRNAs的表达未出现明显变化。在脊髓背角未检测到5-羟色胺5B和5-羟色胺6受体亚型mRNAs的表达。 结论:5-羟色胺受体亚型mRNA在坐骨神经分支选择性损伤模型脊髓背角中的表达具有不同的时程变化特点,提示它们在坐骨神经分支选择性损伤所致的神经病理性痛中发挥不同作用。
AIM: To observe the change of 5-hydroxytryptamine receptor (5-HT4-7) subtype mRNAs expression in the spinal dorsal horn of rat models with spared nerve injury (SNI). METHODS: The experiment was performed in the Department of Anatomy, Fourth Military Medical University from October 2004 to June 2005. ①Twenty-seven SD rats were selected and randomly divided into two groups: normal control group (n = 3) and spared nerve injury (SNI) group (n=24). ②The rats were anaesthetized with barbital sodium (40 mg/kg) and the skin of the lateral left thigh was incised. The cranial and caudal parts of the biceps femoris muscle were separated and held apart by a retractor to expose the sciatic nerve, and the tibial and common peroneal nerves were ligated. The nerves were cut off at 0.5 cm from distal end of deligation in order to establish SNI models. Any stretching or contact with the intact sural nerve was avoided. Sham controls involved exposure of the sciatic nerve and its branches without any lesion on the right thigh. Other process was the same to the operative side. ③At 1, 2, 3, 4, 7, 14, 21 or 28 days after operation, spinal dorsal horns of L3-L6 segnmnts were removed and reverse transcriptase-polymerase chain reaction (RT-PCR) technique was used to detect the changes of 5-HT.7 receptor subtypes mRNAs expression in the spinal dorsal horn. RESULTS: A total of 27 rats were involved in the final analysis.①The expressions of 5-HT4, 5-HT54 and 5-HT7 receptor subtype mRNAs were found in spinal dorsal horn of normal rats, but there were no expressions of 5-HT5B and 5-HT6 receptor subtype mRNAs. ②Up-regnlation of 5-HT4, 5- HT5A and 5-HT7 receptor subtype mRNAs expression was appeared in the operation group. The 5-HT4 receptor subtype mRNA expression significantly increased at the 4^th day after operation, reached a peak at the 21^th day, and then descended to the nomual level at the 28^th day.
