目的观察葛根异黄酮对去卵巢大鼠血管内皮功能的影响及机制。方法雌性SD大鼠行双侧卵巢切除术后,分别灌胃给予150mg/kg和50mg/kg葛根异黄酮,时间为12周。酶联免疫吸附法检测血清雌二醇(E2)和内皮素(ET)含量。硝酸还原酶法检测血清一氧化氮(NO)水平。在分离的各组动脉环标本上,观察乙酰胆碱诱发的内皮依赖性血管舒张反应和硝普钠诱发的非内皮依赖性血管舒张功能变化。Western blot方法观察各组内皮组织样品的内皮型一氧化氮合酶(eNOS)表达变化。结果与假手术组比较,去卵巢组大鼠的血清E2、ET和NO水平出现显著改变,而且其动脉环对乙酰胆碱的舒张功能出现显著性障碍,但对硝普钠的舒张效应无影响。在去卵巢术后给予大小剂量的葛根异黄酮干预后,血清E2、ET和NO水平以及内皮舒张功能,与去卵巢组比较有统计学意义(P〈0.05)。此外,葛根异黄酮也可上调去卵巢大鼠内皮组织的eNOS表达。结论葛根异黄酮具有保护去卵巢大鼠血管内皮功能作用,其机制与类雌激素样作用、上调eNOS表达和降低ET水平等因素有关。
Objective To explore the mechanism and effect of total isoflavones from pueraria lobata(TIP)on vascular endothelial function in ovariectomized rat.Methods The female Sprague-Dawley(SD)rats underwent bilateral oophorectomy were fed 150 mg/kg and 50 mg/kg of TIP for 12 weeks.Serum estradiol(E2)and endothelin(ET)content were measured by enzyme-linked immunosorbent assay,and serum nitric oxide(NO)levels were detected by nitrate reductase assay.Acetylcholine-induced endothelium-dependent vasodilation and sodium nitroprusside-induced reaction were observed in isolated aortic rings in each group rats.Endothelial nitric oxide synthase(eNOS)expression was observed by western blot in the endothelial tissue samples of each group rats.ResultsCompared with the sham group,the serum E2,ET and NO levels in ovariectomized rats had a significant changes,and its arterial rings significantly alter the diastolic function of acetylcholine,but had no effect on the relaxation of sodium nitroprusside.After giving the low and high dose of TIP in ovariectomized rat,there were significant differences in serum E2,ET and NO levels and endothelial function when compared with ovariectomized rats(P〈0.05).In addition,TIP increased the expression of eNOS in endothelial tissue of ovariectomized rat.Conclusion TIP has a role in protecting vascular endothelial function in ovariectomized rat,its mechanism associated with estrogen-like effects,increasing eNOS expression and reduce the level of ET.
