目的:观察丹酚酸B(SalB)预处理的内皮祖细胞(EPCs)不同比例与骨髓间充质干细胞(BMSCs)联合心脏移植后BMSCs的存活状况。方法:通过结扎左冠状动脉复制大鼠急性心肌梗死模型;密度梯度离心法和贴壁筛选法培养、纯化EPCs与BMSCs,免疫细胞化学法鉴定细胞;丹酚酸B最佳药物浓度处理的EPCs以不同比例与5-溴脱氧尿核苷(Brdu)标记的BMSCs混合,移植至心肌梗死区。4周后,硝基四氮唑蓝(N-BT)染色法测算心梗面积;免疫组化法检测Brdu的表达情况。结果:8:1组和模型组心肌梗死面积分别为(9.23±2.35)%和(19.60±3.23)%,8:1组与其他各组比较心梗面积显著缩小(P〈0.05)。4:1组和8:1组BMSCs阳性细胞数与其他治疗组比较,存活细胞显著增多(P〈0.05)。结论:丹酚酸B预处理的EPCs联合BMSCs的移植,能够提高BMSCs在梗死周边和缺血局部的存活。随着EPCs移植比例的增加,BMSCs的存活数量增多。
Objective: To investigate the survival of bone marrow mesenchymal stem cells transplantation in acute myocardial infarction rats,the endothelial progenitor cells preconditioned by salvianolic acid B blend with the bone marrow mesenchymal stem cells on different proportion.Methods: The rat model of AMI was produced by ligation of left anterior descending artery.EPCs and BMSCs in vitro were cultivated and purified by density-gradient centrifugation and plastic adherence methods,and identified by immunocytochemistry.BMSCs marked in Brdu and EPCs preconditioned by the optimum salvianolic acid B were injected into the area of the infarct.After 4 weeks,we examined the infarcted heart area by the N-BT Staining Method and the expression of Brdu by immunocytochemistry.Results: 1.The myocardial infarct area were respectively(9.23±2.35)% and(19.60±3.23)% in 8:1 group and model group.8:1 group was decreased remarkably compared with all groups.2.The positive BMSCs count in 4:1 group and 8:1 group were increased remarkably compared with other treatment groups.Conclusions: EPCs preconditioned with Salvianolic acid B can increase BMSCs Surviving by EPCs combined with BMSCs transplantation.With increasd the proportion of EPCs,the Surviving BMSCs quantity were markedly larger in vivo.
