目的:探讨瑞舒伐他汀对脑缺血再灌注损伤大鼠脑组织中选择素E(E-selectin)和选择素L(L—selee.tin)表达的影响。方法:SD大鼠24只随机分为假手术组(sham)、模型组(model)和治疗组(test)。线栓法制作大鼠大脑中动脉脑缺血再灌注模型,于缺血2h再灌注24h后断头取脑,免疫组织化学法、RT—PCR法和WesternBlot检测脑组织中E.选择素和L-选择素的表达量,结果进行统计学分析。结果:大鼠脑缺血再灌注损伤后,模型组选择素E和选择素表达明显高于假手术组(P〈0.01);与模型组相比,治疗组选择素E和选择素表达明显下调(P〈0.05)。结论:瑞舒伐他汀可能通过下调选择素E和选择素表达而对大鼠脑缺血再灌注损伤发挥保护作用。
Objective: To explore expression of E-selectin and L-selectin affected by rosuvastatin in rats with the cere- bral ischemia-reperfusion. Methods: Rats were randomly assigned into three groups: sham operation group (sham group) , cerebral ischemia-reperfusion group (model group) and E-selectin and L-selectin group (test group). The mid- dle cerebral artery occlusion reperfusion model was made by the suture method ( ischemia for 2 hours, and reperfusion for 24 hours). The level of E-selectin and L-selectin expression was measured by immunohistochemistry, RT-PCR and West- ern Blot. Results: After eerebral ischemia reperfusion injuryin rats, compared with sham group, the expression of E-se- lectin and L-selectin were increased significantly in the model group ( P 〈 0.01 ). Compared with model group, the expres- sion of E-selectin and L-selectin were decreased significantly in the test group ( P 〈 0.05 ). Conclusion: Rosuvastatin can suppress the expression of E-selectin and L-selectin in cerebral isehemia-reperfusion.
