目的:了解蓖麻毒蛋白(Ricin)及从大戟科植物蓖麻种子中提取出来的一种核糖体失活蛋白的热稳定性和抗肿瘤活性。方法:利用亲和层析的方法纯化蓖麻种子毒蛋白,并通过SDS-PAGE电泳和MTT染色法等方法检测其热稳定性、抗肿瘤性。结果:Ricin在37℃的缓冲液中保温60 h,没有明显的降解;能有效地杀死食管癌细胞EC109。结论:Ricin是有前途的蛋白类抗肿瘤药物。
Objective:To study the antitumor activity and the heat stability of Ricin,a kind of the ribosome-inactivating proteins(RIP) isolated from castor bean,euphorbia.Methods:Ricin was purified by affinity chromatography on Sepharose 4B,its heat stability and antitumor activity were respectively detected by SDS-PAGE and MTT method.Results: Ricin can exist in buffer solution for 60 hours without degradation at 37 ℃ and can kill esophageal cancer cells EC109.Conclusion: Ricin is a kind of protein which has a broad prospect of antitumor.
