目的探讨基因与脑胶质瘤的相关性及其表达改变对胶质瘤U87细胞凋亡和增殖能力的影MACC1响。方法荧光定量PCR检测45例脑胶质瘤及相应癌旁组织中MACC1基因的表达;设计并合成基因特异MACC1性的siRNA,转染U87细胞;Western blot检测转染后MACC1蛋白的表达;双标流式细胞法检测细胞凋亡;MTT法测定细胞生长抑制率。结果基因在脑胶质瘤组织中表达明显上调;转染基因特异性的siRNA后MACC1蛋白表达明显降低,凋亡明显增加,生长抑制率明显增加,细胞增殖活力明显降低。结论 MACC1基因的表达异常与脑胶质瘤的发生相关,抑制其表达能促进肿瘤细胞的凋亡同时抑制细胞增殖。
Objective To research the relationship of the expression changes of metastasis-associated in colon cancer-1(MACC1) gene with brain glioma, and the influence of its expression to apoptosis and proliferation in U87 cell. Methods The expression of MACC1 gene was detected in 45 cases of brain glioma and peri-cancerous normal tissues by fluorescent quantitation PCR. Specific siRNA for MACC1 gene was designed and synthesized, and then U87 cells were transfected with siRNA-MACC1 to inhibit the expression of MACC1 gene. After RNA interference, the expression of MACC1 protein was detected by western blot, the apoptosis of the transfected cells was detected by flow cytometry and the inhibition ratio of cell proliferation was detected by methyl thiazolyl tetrazolium (MTT). Results The expression of gene presented an obvious increasing in brain glioma. After RNA interference to MACC1 gene, the expression of MACC1 protein in the transfected U87 cells was downregulated significantly, however, the apoptosis rate and the inhibition ratio of cell proliferation in the transfected U87 cells increased significantly. Conclusions The overexpression of MACC1 gene in brain glioma might be related to the pathogenesis of brain glioma, inhibition of MACC1 gene can increase apoptosis of U87 cells and inhibit the proliferation of tumor cells.
