利用高效液相色谱-电喷雾-飞行时间质谱(HPLC—ESI—TOF/MS)在负离子模式下成功检测了荼籽粕中提取的茶皂素。高效液相色谱分析茶皂素超纯品得到5个主要皂苷峰,出峰时间分别为4.01、19.52、27.74、29.23和34.01min,各吸收峰峰形对称,由此可见茶皂素组分在该条件下能够得到较好分离。不同浓度荼皂素的定量曲线表明5个峰的峰面积与茶皂素含量的线性回归方程相关系数r值均在0.99以上。ESI—TOF/MS质谱分析结果表明5个组分分别含有2~5种五环三萜类皂苷,合计18种。该法有较高的精确性,可用于茶皂素的定性、定量分析。
Tea Saponin in tea seed meal hads been successfully separated and identified by high performance liquid chromatography - quadrupole/time of flight mass spectrometer ( HPLC - TOF/MS ). The eluted times of Tea Saponins were respectively 4.01,19.52,27.74,29.23 and 34.01 min,, and the peak shapes were symmetrical and with good resolving power. The linearity of each standard curve was confirmed by plotting of the peak area and matc- hing concentration of the Tea Saponins solution. All the correlation coefficient values of linear regression equations for the five components were more than 0.99. The results of TOF/MS analysis showed that there were 2 to 5 triterpenoid Saponin ingredients in 5 separated components,and there were 18 Saponins in total. The results showed a rather precise method, which could be used for quantitative and qualitative analysis of Tea Saponin.
