利用Plackett—Burman试验设计从8种营养成分中筛选出4种对多杀菌素产量影响较显著的因子:葡萄糖、棉籽粉、玉米浆、豆饼粉,而后通过D-最优混料设计,对这4种成分进行优化设计。以多杀菌素的产量为期望,进行回归分析并建立关于多杀菌素产量的二阶拟合方程,确定4种成分的最佳比例为:葡萄糖59g/L、棉籽粉28g/L、玉米浆6.5g/L、豆饼粉6.5g/L,根据方程预测产量理论最高值为256.75mg/L,按此配方进发酵验证,产量为249.9mg/L,证明该方程的拟合性较好。
Glucose, cotton seed flour, corn steep liquor and soybean flour were found to be the most important factors affecting spinosad production using Plackett-Burman design from 8 factors. Then, Doptimal mixture design to optimize the 4 factors. Regression model on Spinosad production was established, and a second-order polynomial model which expressed the good relationship of variables and response. The optimum fermentation medium for spinosad production were as follows: Glucose 59 g/L, cotton seed flour 28 g/L, corn steep liquor 6.5 g/L, soybean flour 6.5 g/L, the predicted yield was 256.75 mg/L. The yield of spinosad was 249.9 mg/L using the optimized medium, and this proved the equation fitted the yield of spinosad accurately.
