目的研究人脑胶质瘤标本中脂肪酸合成酶(FASN)的表达水平及其临床意义。方法体外常规培养人脑胶质瘤细胞系U87和U373,免疫荧光染色检测U87和U373细胞FASN的表达。收集首都医科大学附属北京天坛医院神经外科自2012年3月至2013年3月手术切除的原发性胶质瘤标本67例,其中WHO分级Ⅰ级3例,Ⅱ级29例,Ⅲ级22例,Ⅳ级13例。免疫组化染色检测胶质瘤标本中FASN、增殖因子Ki-67、CD34的表达,比较不同级别胶质瘤标本中FASN、Ki-67的表达水平及微血管密度(MVD)。Spearman相关分析胶质瘤标本FASN与Ki-67表达水平、FASN表达水平与MVD之间的关系。结果免疫荧光染色检测显示U87和U373细胞浆中均表达FASN;免疫组化染色显示人胶质瘤标本中FASN定位于细胞浆,Ki-67定位于细胞核,CD34定位于细胞膜。Ⅰ级胶质瘤标本中FASN的表达(3.3683±0.6549)低于Ⅱ级(4.0512±0.4859),Ⅲ级胶质瘤标本中FASN的表达(4.1881±0.5755)低于Ⅳ级(4.6996±0.5164),差异有统计学意义(P〈0.05);Ⅱ级胶质瘤标本MVD低于Ⅲ级,Ⅲ级胶质瘤标本MVD低于Ⅳ级,差异有统计学意义(P〈0.05);11级胶质瘤标本Ki-67的表达低于Ⅲ级,差异有统计学意义(P〈0.05)。Spearman相关分析显示胶质瘤标本中FASN与Ki-67的表达呈正相关关系(r=0.636,P=0.014),且FASN的表达与MVD也呈正相关关系(r=0.606,P=0.030)。结论人脑胶质瘤中FASN的表达水平越高,肿瘤细胞的增殖能力越强,血供趋于越丰富,肿瘤的恶性度趋于增高。
Objective To investigate the expression level of fatty acid synthase (FASN) in different grades of gliomas and its relationship with glioma malignancy. Methods Glioma U87 and U373 cell lines were routinely cultured in vitro; immunofluorescence assay was performed to detect the cellular localization of FASN in glioma U87 and U373 cell lines. Sixty-seven human glioma samples, collected in our hospital from March 2012 to March 2013, were used in our study; grade I was noted in three samples by WHO grading, grade Ⅱ in 29, graded Ⅲ in 22, and grade Ⅳ in 13; immunohistochemistry was performed to detect the expression levels of FASN, CD34 and Ki-67 and microvascular density (MVD) in these human glioma samples. Spearman's correlation was used to analyze the relationship between FASN expression and both Ki-67 expression and MVD in human gliomas. Results (1) FASN could express in the cytoplasm of glioma U87 and U373 cell lines. (2)immumohistochemical staining indicated that FASN mainly located in the cytoplasm, Ki-67 in the cell nucleus, and CD34 in the cytomembrane. FASN expression in samples of grade Ⅰ (3.3683 ±0.6549) was significantly lower than that in samples of grade Ⅱ (4.0512±0.4859, P〈0.05), and FASN expression in samples of grade Ⅲ (4.1881±0.5755) was significantly lower than that in samples of grade Ⅳ(4.6996± 0.5164, P〈0.05); MVD in samples of grade II was significantly smaller than that in samples of grade Ⅲ (P〈0.05), and MVD in samples of grade Ⅲwas significantly lower than that in samples of grade IV (P〈0.05); Ki-67 expression in samples of grade Ⅱ was significantly lower than that in samples of grade HI (P〈0.05). (3) There was a positive and significant correlation between FASN expression and MVD (r=0.606, P=-0.030); a positive and significant correlation between FASN expression and Ki-67 was also noted (r=0.636, P=O.014).
