目的研究黄连(CoptischinensisFranch.)根茎的化学成分。方法利用硅胶柱色谱、凝胶柱色谱、中低压液相色谱、高效液相色谱等色谱技术对黄连的化学成分进行分离和纯化,通过NMR等波谱数据分析确定化合物的结构。结果从黄连正丁醇层提取物中分离得到11个已知成分,分别鉴定为降氧化北美黄连次碱(noroxyhydrastineine,1)、3,4-二氢-6,7-二甲氧基异喹诺酮(6,7-di-methoxy-3,4-dihydro-2H-isoquinolin-1-one,2)、8-氧化黄连碱(8-oxocoptisine,3)、小檗碱(berberine,4)、氧化小檗碱(oxyberberine,5)、原儿茶酸甲酯(protocatechuicacidmethylester,6)、丹参素甲正丁酯(n-butyl 3,4-dihydroxyphenyllactate,7)、反式-3,4-二甲氧基肉桂酸(E-3,4-dimethoxycinnamicacid,8)、阿魏酸正丁酯(n-butyl ferulate,9)、5-O-feruloylquinicacidbutylester(10)、4-O—feru—loylquinic acid butyl ester(11)。结论化合物7~11为首次从黄连属植物中分离得到。
Objective To study the chemical constituents of rhizoma of Coptis chinensis. Methods The com- pounds from the n-butanol extracts were isolated by silica gel column, Sephadex LH-20 column chromatography and HPLC techniques. And these compounds were identified by the physico-chemical properties and spectroscopic methods. Results Eleven compounds were isolated from the n-butanol extract of C. chinensis, whose structures were determined as noroxyhydrastineine( 1 ) ,6,7-dimethoxy-3,4-dihydro-2H-isoquinolin-1- one ( 2 ), 8-oxocoptisine ( 3 ), berberine ( 4 ), oxyberberine ( 5 ), protocatechuic acid methyl ester ( 6 ), n -bntyl3, 4-dihydroxyphenyllactate ( 7 ), E-3,4-dimethoxycinnamic acid ( 8 ), n-butyl ferulate ( 9 ), 5-O-feruloylquinic acid butyl ester(10) ,4-O-feruloylquinic acid butyl ester( 11 ). Conclusions Compounds 7-11 are isolated from C. chinensis for the first time.
