目的观察汉滩病毒(HTNV)感染的TLR4基因沉默的EVC304细胞(TLR4-EVC304)转录因子NF-κB和IRF-3的细胞核移位情况,为抗HTNV固有免疫及其信号转导研究提供新资料。方法用汉滩病毒76.118株分别感染TLR4-和TLR4+EVC304细胞,同时以LPS作为阳性对照组,无任何刺激作为阴性对照组,6h后用间接免疫荧光方法检测NF—κB和IRF-3的细胞核移位现象。结果汉滩病毒76—118株刺激6h后,在TLR4+EVC304细胞中,NF—KB和IRF-3发生细胞核移位,而在TLR4-EVC304细胞中未出现细胞核移位现象。结论TLR4可能介导了HTNV感染的EVC304细胞中NF—κB和IRF-3的细胞核移位。
Objective To observe the nuclear translocation of transcription factor NF-κB and IRF- 3 in TLR4 silenced EVC304 ceils infected by HTNV and to provide new information for anti-HTNV innate immunity and its signal transduction. Methods TLR4 - cells and TLR4 + cells were infected by HTNV 76- 118, respectively. The cells stimulated by LPS were selected as positive control groups, and the cells without stimulation were selected as negative control gronps. After 6 hours, indirect immanofluorescence assay(IFA) was used to detect the nuclear translocation of NF-κB and IRF-3. Results The transcription factor NF-κB and IRF-3 transfered into nuclear 6 hours after stimulated by HTNV 76-118. Conclusion TLR4 may medi- ate the nuclear translocation of transcription factor NF-KB and IRF-3 in HTNV infected human umbilical vein endothelial cells.
