目的:比较两种制作大鼠急性心肌梗死模型的冠状动脉结扎法,推荐一种稳定性高、生存率高的模型制作方法。方法:将40只SD大鼠按照随机数字法分为两组:常规组和改进组,比较两组的手术时间、生存率,术后24h测量两组大鼠心肌梗死面积,危险区面积以危险区占左室面积的百分比(Ar/LV)表示,心肌梗死面积用梗死区占危险区面积的百分比(An/Ar)表示,比较两组Ar/LV和An/Ar。结果:术后3h改进组和常规组的存活率分别为90%和60%,二者比较无统计学差异(P〉0.05);术后24h改进组大鼠的存活率为85%,常规组为45%,两组间有显著的统计学差异(P〈0.05);常规组Ar/LV和An/Ar分别为51.4%±2.5%和52.5%±2.4%,改进组Ar/LV和An/Ar分别为51.5%±2.3%和52.5%±2.2%,两组间均无统计学差异(P〉0.05);常规组的操作时间明显长于改进组,为(627.9±16.0)s对(253.9±12.7)s(P〈0.01)。结论:改进的模型制作方法提高了大鼠生存率,缩短了手术时间,具有显著优越性,值得推广。
Objective:Through comparing two kinds of acute myocardial infarction models generated by coronary artery ligation,to recommend a stable,repeatable and long-lasting rat model of acute myocardial infarction. Methods:40SD rats were randomly divided into two groups:conventional group and modified group.The operation time,survival rate were compared between two groups.After24 hours,the myocardial infarction area of the two groups was compared.The area at risk was represented as the percentage of risk area against left ventricular area(Ar/LV),and the area of necrosis was represented as the percentage of necrosis area against risk area(An/Ar). Results:After 3hours,the survival rate in the modified group was 90%,while in the conventional group it was 60%,with no statistical significance between the two groups(P〉0.05);After 24 hours,the survival rate in the modified group was 85%,while in the conventional group it was 45%,showing statistical significance between the two groups(P〈0.05);The Ar/LV and An/Ar in the conventional group were 51.4% ±2.5% and 52.5%±2.4%,respectively,while the Ar/LV and An/Ar in the modified group were 51.5%±2.3% and 52.5%±2.2%,respectively,indicating that there was no statistical significance in Ar/LV or An/Ar between the two groups(P〉0.05);The operating time in the conventional group was(627.9±16.0)s,while in the modified group it was(253.9±12.7)s,showing statistical significance between the two groups(P〈0.05). Conclusion:The modified model method can greatly improve survival rates,shorten surgical time and offers significant superiority to the conventional methods.It is worth recommending.
