目的:筛选能与炎症相关趋化因子结合的广谱抑制剂先导物。方法:用内毒素刺激外周血单核细胞建立急性炎症模型,以刺激炎症相关趋化因子的产生,利用噬菌体随机12肽库进行高通量筛选得到阳性克隆,并初步分析其体外活性。结果:经过四轮肽库亲和筛选后,获得了一共同短肽(TVVGSGCVLRIQ)。结论:该短肽可有效抑制炎性趋化因子活性,说明该短肽有可能成为一种广谱的炎性趋化因子抑制先导物。
The aim of the study is to screen the broad-spectrum inflammatory chemokines inhibitor leader. We stimulated peripheral blood mononuclear cells (PBMC) with LPS to set up an acute inflammation model and to produce the inflammatory chemokines. Then by using the phage random peptide library, the positive clones were screened and their in vitro activity was analyzed. At last we found a common short peptide (TVVGSGCVLRIQ) by four screening rounds; the short peptide can effectively inhibit the inflammatory chemokines activity. All the results indicate that the peptide may be a broad spectrum of inflammatory chemokine inhibitors leader.
