目的观察中药远志YZ-50对慢性应激抑郁模型大鼠行为学和血清促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、血清皮质醇(COR)含量的影响。方法选择Open-filed法评分相近的♂wistar大鼠100只分为空白对照组、模型组、地昔帕明(20mg·kg^-1)、远志YZ-50低、高剂量(2.8、5.6g生药/kg),每组20只。选用慢性轻度不可预见性应激结合孤养造模,观察各组大鼠敞箱实验、体重以及糖水摄入等行为学指标变化,采用放射免疫方法检测大鼠血清中CRH、ACTH和COR含量。结果慢性应激抑郁大鼠体重增加缓慢,敞箱实验中的水平运动、垂直运动得分、糖水消耗明显下降,而且其血清CRH、ACTH和COR含量显著增加。地昔帕明组和远志YZ-50低、高剂量组均能有效改善慢性应激抑郁大鼠模型的行为学和神经内分泌变化。结论慢性轻度不可预见性应激结合孤养造模可使大鼠行为学及神经内分泌发生异常改变,引起抑郁状态,远志YZ-50对此具有不同程度的拮抗作用,发挥出一定的抗抑郁作用。
Aim To observe the effect of Polygala Tenuifolia Willd YZ-50 on neuroendocrine and behavior of chronic stress depression rats. Methods one hundred male wister rats were randomly divided into five groups : normal control group, model group, desipramine group (20mg·kg^-1) and Polygala Tenuifolia Willd YZ-50 low (2.8g dried medicinal herb/kg)and high(5.6g dried medicinal herb/kg)groups. All rats except the control group were singly housed and exposed to an unpredicted sequence of mild stressor. The behavior of rats was detected by open - field test weight and sucrose preference test , and the levels of CRH ( corticotrophin releasing hormone), adrenocorticotropic hormone (ACTH) and cortisol (COR) in blood serum in different groups were observed by RIA. Results Compared with normal control group , the rats in the model group showed decreased body weight ,lower sucrose water consumption test, reduced crossing and rearing scores significantly. The concentration of blood serum CRH, ACTH and COR increased obviously in stress - model rats compared to control groups. Results also demonstrated that desipramine. Polygala Tenuifolia Willd YZ-50 low and high dosage groups can improve behavioral disorder and reduce the levels of blood serum CRH, ACTH and COR. Conclusion Chronic unpredicted mild stress can affect rats' neuroendocrine and behavior and cause depression. Polygala Tenuifolia Willd YZ-50 has antagonism against it and has some anti-depressant effect .
