采用液体深层发酵的方法,研究了培养条件对棘豆蠕孢菌(Embellisia oxytropis)菌丝体生长的影响,并采用L9(3^4)正交试验优化液体发酵培养基组成。结果表明,试验范围内棘豆蠕孢菌液体发酵培养的最佳条件为:培养基初始pH6.5、培养温度24℃、装液量32%、接种量12%,在此条件下100mL发酵液的菌丝体产量为0.573g;优化培养基配方为:玉米粉5%、马铃薯20%、磷酸二氢钾0.1%、硫酸镁0.08%。菌种扩繁次数对液体发酵的影响较大,第3代开始生物量显著下降。
The effects of submerged-culture conditions on the mycelium growth of Embellisia oxyt- ropis were studied,and the medium composition was optimized by the L9 (3^4)orthogonal experimental method. The results showed that the optimal cultural conditions were pH 6.5 at initial stage, culture temperature of 24 ℃ ,medium volume of 32% flask capacity and pitching rate of 12% (v/v),in which the mycelium yield of 100 mL fermented broth was 0. 573 g;and the optimum medium composition was 5% corn meal,20% potato,0.1% KH2PO3 and 0.08% MgSO4. The frequency of subculture had a great effect on the mycelium yield which declined significantly after the second transfer.
