目的研究Atarora—A在宫颈癌中的表达情况,以及其表达水平与宫颈癌预后的关系。方法选择2007年1月至2009年1月第四军医大学西京医院妇产科宫颈鳞状细胞癌与宫颈腺癌患者组织标本共74例,对照组选取同期保存的该患者癌旁正常组织(经病理检查证实无肿瘤细胞扩散),通过半定量RT.PCR、蛋白印迹实验在细胞学水平对Aurora-A的表达进行检测;通过免疫组化、实时定量PCR的手段明确Aurora—A在宫颈癌及正常癌旁组织中的表达情况;在此基础上将其表达水平与患者临床病理特征进行统计学分析。结果半定量RT—PCR与Western—blotting实验结果均提示肿瘤细胞系中Aurora—A的表达明显高于正常细胞;实时定量PCR提示51.35%(38/74)的组织标本中Aurora-AmRNA由0.088±0.102升高至0.623±0.410(P〈0.001),并且与患者的FIGO分期、肿瘤细胞分化水平、是否存在宫旁浸润、是否存在远处转移等因素呈正相关关系。结论Aurora.A在宫颈癌组织中存在高表达状态,并且与宫颈癌预后之间存在密切关系。
Objective To observe the expression level of Aurora-A in cervical tissues and cells, and investigate whether Aurora-A expression correlates With clinicopathologic factors and prognosis of cervical carcinoma patients. Methods Dif- ferential expression of Aurora-A was examined by RT-PCR,Western bloting and Immunohistochemistry analysis in differ- ent cervical tissues and cell lines;to analyze whether Aurora-A expression correlates with clinicopathologic factors and prognosis of cervical carcinoma patients with statistics. Results Aurora-A is over expressed in human cervical cancer, just like the condition in other cancers, and the overexpression of Aurora-A is associated with FIGO stage, tumor differenti- ation, parametrial invasion and lymphnode or hematogenous metastasis, but not other clinicopathological factors. Conclusion Aurora-A over expresses in cervical cancer, and the level of expression correlate with the prognosis of cervical cancer patients.
