目的 分离纯化退黄藤中的东莨菪内酯并测定其含量。方法 采用硅胶柱色谱分离纯化退黄藤中的东莨菪内酯,并根据理化性质及光谱数据鉴定其结构;采用HPLC法测定其含量,色谱柱为Zorbax Eclipse XDB-C(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%冰乙酸(39∶61),流速1.0 m L·min^-1,检测波长345 nm。结果 东莨菪内酯3.11-31.1μg·m L^-1与峰面积呈良好的线性关系(r=0.9999),平均回收率为99.29%,RSD=1.40%(n=6)。结论 所用方法简便、准确、快速,可用于退黄藤药材的质量控制。
OBJECTIVE To isolate,purify and determine the scopoletin in Pegia sarmentosa ( Lecte. ) Hand. - Mazz.. METHODS Scopoletin was isolated from P. sarmentosa by the silica gel column chromatography,and its structure was identified on the basis of spectral characteristics. Zorbax Eclipse XDB - C18 (250 mm×4.6 mm,5 μm) reversed - phase column was used with mobile phase as methyl alcohol -0. 1% acetic acid solution(39 : 61 ). The flow rate was 1.0 mL·min^-1 with the detection wavelength of 345 nm. RESULTS The linear range for scopoletin was 3.11 -31.1 μg·mL^-1 (r =0. 9999). The average recovery was 99.29% with the RSD of 1.40% (n = 6). CONCLUSION The method is simple,accurate and rapid,and it can be used for quality control of P. sarmentosa.
