目的:建立大鼠实验性牙髓炎模型,了解Cryopyrin在大鼠牙髓组织的表达情况。方法:30只Sprague—Dawley大鼠随机分为0,1,3,5,7d5组。除0d组外,其余四组大鼠双侧下颌第一磨牙开髓后暴露于VI腔环境中,分别于开髓后1,3,5和7d处死大鼠,分离双侧下颌骨。常规组织学处理后HE染色观察牙髓炎症状况,免疫组化检测Cryopyrin在牙髓炎中的表达及定位。结果:大鼠正常牙髓组织成牙本质细胞层Cryopyrin阳性表达,牙髓内部血管内皮细胞偶见表达,牙髓成纤维细胞未见表达。炎性牙髓组织血管内皮细胞表达增加,牙髓成纤维细胞及多种炎症细胞均有阳性表达,且表达的数量和牙髓的炎症浸润强度显著正相关(r=0.655,P〈0.01)。结论:Cryopyrin在牙髓组织中表达并在牙髓炎的发生发展中起着重要作用。
To investigate the expression and localization of Cryopyrin in the development of experimental pulpitis in rat. Methods: Thirty Sprague-Dawley rats were randomly assigned into five groups according to the excution day. The pulps of bilateral mandibular first molar were ex- posed in the oral environment except the rats of day 0 group. The animals in the other groups were sacrificed at on the 1st, 3rd, 5th and 7th days respectively after pulpal exposure. The bilat- eral mandibles samples were extracted for histological processing, then they were stained by he- matoxylin-eosin (HE) to examine inflammation infiltration in the pulp, as well as immunohisto- chemcally examined for the Cryopyrin. Results: Cryopyrin was detected in the cytoplasm of odon- toblasts and vascular endothelial cells, but not in the fibroblasts of the normal pulp. In the in- flammatory pulp tissue, fibroblasts, monocytes, neutrophils, macrophages and vascular endothe- lial cells all expressed Cryopyrin. The number of Cryopyrin-positive cells was significantly corre- lated with the intensity of inflammatory infiltration (r=0. 655, P〈0.01). Conclusion: Cryopyrin is expressed in the normal and inflammatory pulp tissues, and plays an important role in the de- velopment of pulpitis.
