目的分析解毒化瘀中药组方水煎液乙酸乙酯分离部分化学组分。方法采用高效液相色谱-质谱联用(HPLC-MS)法,以甲醇-水为流动相,梯度洗脱,流量为1mL·min-1,采用ESI正离子模式进行质谱检测,相对分子质量扫描范围为50~1 000m/z。将检测得到的相对分子质量数据与Aglient中药成分数据库比对,对相似度得分在80%以上的化合物进行文献及来源药材成分检索,鉴定化合物。结果共鉴定出2-O-桂皮酰基-β-葡萄糖、淫羊藿属甙C、大黄二蒽酮A、欧芹脑、9,10-二甲氧基紫檀烷-3-O-β-D-葡萄糖甙、葛缕酮等18种化合物,2-O-桂皮酰基-β-葡萄糖、大黄二蒽酮A、大黄酚来源于大黄;9,10-二甲氧基紫檀烷-3-O-β-D-葡萄糖甙、异微凸剑叶莎醇、熊竹素来源于黄芪;欧芹脑、葛缕酮、茵陈蒿B来源于茵陈;淫羊藿新甙C、1,2-双(4-羟基-3-甲氧基苯基)-1,3-丙二醇来源于淫羊藿。7,9(11)-去氢茯苓酸、藜芦醇、赛仙鹤草酚C分别来源于茯苓、虎杖、仙鹤草;苍术内酯I来源于白术;决明萘乙酮-8-O-β-D-葡萄糖甙来源于虎杖和大黄;槲皮素来源于仙鹤草、淫羊藿、茵陈。结论采用HPLCMS法可明确中药组方水煎液乙酸乙酯分离部分的化学组分,为解毒化瘀中药组方的临床应用提供了理论依据。
Objective To analyze the chemical constituents of sepeartion part of ethylacetate in water-decocted liquid of JIEDU HUAYU traditional Chinese medicine. Methods Applying HPLC-MS method,taking methanol-water as mobile phase,gradient elution with velocity of flow as 1mL·min-1,mass-spectrometric detection was done using ESI positive ion mode,scanning range of relative molecular mass was 50-1 000m/z.The detected relative molecular mass data were compared with Aglient traditional Chinese medicine composition database.For compounds with their score of similarity of more than 80% were selected for literature retrieval and source of medicinal herbs identification. Results A total of 18compounds-2-O-cinnamoyl-β-glucose,epimedoside C,reidin A,apiole,9-,10-dimethoxypterocarpan-3-O-β-D-glucoside,carvone,2-O-cinnamoyl-β-glucose,chrysophanol,reidin A,and so on-were identified.2-O-cinnamoyl-β-glucose,reidin A and chrysophanol originated from rheum;9-and10-dimethoxypterocarpan-3-O-β-D-glucoside,isomucronulatol,and kumatakenin originated from astragalus membranaceus;apiole,carvone and capillartemisin B from capillary artemisia;Epimedoside C and 1,2-bis(4-hydroxy-3-methoxyphenyl)propan-1,3-diol from epimedium.7,9(11)-dehydropachymic acid,resveratrol,and agrimol C respectively originated from poria cocos,poly-gonum cuspidatum and agrimonia pilosa.Atractylenolide I originated from atractylodes macrocephala koidz;Torachrysone 8-O-β-D-glucopyranoside from rheum officinale and polygonum cuspidatum;quercetin originated from agrimonia pilosa,epimedium and ca-pillary artemisia. Conclusion Employing HPLC-MS technology,the chemical constituents of separation part of ethylacetate in water extract of JIEDU HUAYU traditional Chinese medicine can be identified,which provides theory basis for clinical application of this prescription.
