目的探讨血府逐瘀汤抗动脉粥样硬化(AS)的作用及其机制。方法将40只SD大鼠随机分为对照组、模型组、血府逐瘀汤组、血脂康组各10只,对照组给予正常饲料,模型组给予高脂饲料,血府逐瘀汤组和血脂康组在给予高脂饲料时给予相应药物。12周后处死大鼠,取主动脉,免疫组化法观察基质金属蛋白酶2(MMP-2)表达情况,全自动生化分析仪检测高敏C-反应蛋白(hs-CRP)水平,酶联免疫吸附(ELISA)法检测单核细胞趋化因子-1(MCP-1)。结果模型组MMP-2、hs-CRP和MCP-1表达水平较对照组明显升高(P〈0.05),血府逐瘀汤组和血脂康组MMP-2、hs-CRP和MCP-1表达较模型组明显降低(P〈0.05),血府逐瘀汤组和血脂康组差异无统计学意义。结论血府逐瘀汤能调节和改善血管弹性,抑制炎症因子表达,防治AS,其机制可能与抑制MMP-2、降低hs-CRP和MCP-1有关。
Objective To investigate the effect and mechanism of xuefu zhuyu decoction on rats with atherosclerosis. Methods Forty SD rats were randomly divided into the control, model, x~fu zhuyu and xuezhikang groups, with 10 rats in each. The group was fed with normal diet, the model was fed with high fat diet, while the treatment groups were fed with high fat diet and drugs. After 12 weeks, the rats were sacrificed. The expression of MMP-2 protein in the endarterium of AS rats was detected by immunohistochemical technique, the level of hs-CRP was detected by full automatic biochemical analyzer, the MCP-1 was detected by ELISA method. Results The level of MMP-2, hs-CRP and MCP-1 increased significantly in the model group (P〈0.05) , but decreased in the treatment group(P〈0.05) . Conclusion Xuefu zhuyu decoction can improve elasticity of blood vessels and inhibit expression of inflammatory factor so as to prevent and treat atherosclerosis, the mechanism of which may be associtated with decreasing the expression of MMP-2, hs-CRP and MCP-1.
