目的:研究载脂蛋白E(ApoE)和原肌球蛋白调节蛋白1(Tmod1)基因双敲小鼠的血液流变学指标和血脂的改变。方法将心脏过表达而全身其他部位敲除Tmod1的小鼠(TOT/Tmod1-/-)与ApoE-/-小鼠杂交获得ApoE和Tmod1的双敲小鼠(ApoE-/-/TOT/Tmod1-/-),并进行基因型鉴定。从ApoE-/-和双敲小鼠取血检测血细胞计数、血液流变学指标和血脂水平。结果同ApoE-/-小鼠相比,双敲小鼠血细胞计数部分指标有显著性改变;红细胞的最大变形指数和变形曲线积分面积明显降低;渗透脆性显著变差;血脂中甘油三酯和高密度脂蛋白胆固醇有显著升高。结论 Tmod1的敲除能够引起小鼠红细胞流变性和血脂水平的改变。
Objective To study the changes of hemorheological indexes and blood lipid in apolipoprotein E (ApoE) and tropomodulin 1 (Tmod1) double gene knockout mice.Methods The Tmod1 gene knockout mice with heart specifi c Tmod1 overexpressing (TOT/Tmod1-/-) were crossbred with ApoE-/- mice to obtain the ApoE and Tmod1 double knockout mice (ApoE-/-/TOT/Tmod1-/-). The mice were genotyped and blood was taken and subjected to blood counting test, hemorheological tests and blood lipid measurements. ResultsCompared with ApoE-/- mice, double knockout mice had changes in some parameters in blood counting test; signifi cantly lower red blood cell deformation index and integrated deformation index; impaired osmotic fragility; and higher blood triglyceride and high density lipoprotein cholesterol.Conclusion The knockout of Tmod1 could cause the changes of hemorheological indexes and blood lipids in mice.
