目的:比较人脂肪基质细胞(human adipose-derived mesenehymal stem cells,hADSCs)在矿化条件与非矿化条件下分别培养,评价矿化液对其增殖和分化的影响。方法:酶消法原代培养人脂肪基质细胞,再以含地塞米松、β-甘油磷酸钠和维生素C的矿化液进行诱导,MTT检测矿化液对细胞增殖的影响,细胞培养上清液检测碱性磷酸酶(ALP)活性及骨钙素(OCN)含量。结果:矿化液抑制细胞的增殖,提高人脂肪基质细胞ALP活性和骨钙素的含量。结论:人脂肪基质细胞在矿化条件下可以表现出成骨细胞特性。
Objective:Toexpl oreosteogenic differentiation of human adipose-derived mesenehymal stem cells bymineralization solution. Methods: Human liposuction aspirates were proteolytically digested and centrifuged to obtain hADSCs. After primary culture in control media and expansion to four passages, the cells were cultured in osteogenic media and control media for 4 weeks respectively.
