目的探讨金属硫蛋白1X(MT1X)在肺癌耐药细胞株(A549/DDP)顺铂耐药中的作用。方法通过化学合成针对MT1X的siRNA段抑制其在A549/DDP中的表达,实时定量聚合酶链反应(PCR)及WB检测干扰效果,M1Tr及平板克隆形成实验检测干扰前后细胞对顺铂的药物敏感性变化;流式细胞术检测细胞周期和细胞凋亡情况。结果针对MT1X的siRNA片段能在RNA及蛋白水平有效抑制其在A549/DDP中的表达;与对照组相比,MT1X表达被抑制后,肿瘤细胞对顺铂的敏感性明显升高;增殖被抑制,平板克隆形成率明显降低(P〈O.05),细胞凋亡率显著增高(P〈0.05)。结论针对MT1X的siRNA片段可部分逆转肺癌细胞(A549/DDP)对顺铂的耐受,为提高肺癌的化疗效率提供了一种新方法。
Objective To investigate the role of metallothionein 1X (MT1 X) in cisplatin-resistant lung cancer cell lines (A549/DDP). Methods The specific MT1X small interfering RNA was synthesized in vitro and transfected into A549/DDP cells. Then, the expression of MT1X was detected by real-time PCR and Western blot, the cisplatin drug sensi- tivity and cell proliferation ability were investigated by MTF and colony formation assay and the cell cycle and apoptosis were analyzed by flow cytometry. Results The results indicated that our MT1 X-interfering RNA successfully diminished MT1X expression at both mRNA and protein level. Suppressing MT1X expression could significantly increase the sensitivity of tumor cells to cisplatin, decrease colony formation and increase the apoptosis of A549/DDP ceils to cisplatin (P 〈 0. 05 ). Conclusion The results suggest that MTl X siRNA can reverse the cisplatin resistance of human lung cancer cell line, and this finding may provide a new method to improve the efficiency of lung cancer chemotherapy.
