αv整合素介导β-淀粉样蛋白诱导的长时程增强的抑制

在新鲜分离大鼠的海马切片上,采用电生理方法记录兴奋性突触后电位,观察整合素在Aβ抑制长时程增强作用的介导机制.研究表明:在离体海马齿状回上,αv整合素的特异性抗体阻止了Aβ对LTP的抑制,而2种对照抗体并没有此作用.此外,非肽类的小分子物质SM256、整合素拮抗配体超纤维连接蛋白、鬼笔环肽和1种从蛇血清分离的小分子蛋白锯鳞蝮蝰血抑环肽也都能阻断Aβ对LTP的抑制.这些研究都表明在阿尔茨海默病神经变性前,州整合素可能是突触可塑性中重要的介质.

英文摘要:

In the present studies, we have investigated the role of integrins in the given Aβ-mediated block of LTP in the dentate gyrus in vitro. Selective antibodies to the αv integrin subunit are found to be able to curb the Aβ inhibition of LTP in the dentate gyrus in vitro. In contrast, two antibodies does not produce such an action of Aβ. In addition, a small molecule nonpeptide antagonist of αv-containing integrins and two other antagonistic ligands of integrins, superfibronectin and the disintegrin echistatin, also show the preventative property of the Aβ inhibition of LTP. The results of the studies indicate that αv integrins may act as the important medias of synaptic dysfunction prior to neurodegeneration in Alzheimer's disease.
