目的探讨金属蛋白酶解离素28(ADAM28)反义核酸对体外培养的小鼠牙胚发育的影响。方法建立Balb/c小鼠牙胚体外培养模型,利用ADAM28AS—ODN抑制小鼠牙胚中ADAM28mRNA的翻译,采用组织学和Western blot检测该抑制反应对牙胚发育的影响。结果AS—ODN组部分成牙本质细胞出现坏死崩解、排列紊乱,髓腔内胶原纤维分泌较少,牙胚组织巾ADAM28的表达明显减弱。结论ADAM28反义核酸可显著抑制发育牙胚的间充质细胞的增殖、成熟与分化,有效封闭牙胚组织中ADAM28的表达。
Objective To investigate the effects of ADAM28 (Adisintegrin and metalloproteinase 28) AS-ODN on development of mouse tooth germ cultured in vitro. Methods Balb/c mouse tooth germ culture model in vitro was constructed and AS-ODN was used to inhibit the translation of ADAM28 mRNA in mouse tooth germ. Histology and Western blot technique were used to detect the effects of its inhibition on tooth germ development. Results Partial odontoblasts of AS-ODN group appeared necrosis, disintegration, confused alignment, and collagen fibers secretion in dental cavity decreased. The expressions of ADAM28 in tooth germ tissue weakened obviously. Conclusion ADAM28 AS-ODN could significantly inhibit the proliferation, maturity and differentiation of mesenchymal cells in developmental tooth germ, and effectively block the expressions of ADAM28 in tooth germ tissue.
