目的:研究青藤碱口服给药在兔生理状态下血液及关节液的药动学特征。方法:经口灌胃给予新西兰兔青藤碱100mg·kg~(-1),采用微透析技术采集关节腔透析液,并同步耳缘静脉采集血液,采用LC/MS/MS法测定血液及关节液中青藤碱浓度。利用DAS2.1.1软件计算药动学参数。结果:青藤碱口服给药后在新西兰兔血液中主要药动学参数Cmax为2 867.5±246.76 ng·mL^(-1),T1/2Ka为0.31±0.10 h,Ka为3.72±2.4 h^(-1),T1/2为3.27±0.50 h,符合一室模型;关节腔中主要药动学参数Cmax为1 874.688±477.19 ng·mL^(-1),Ka为3.6±0.75 h^(-1),T1/2为3.91±1.48 h,符合一室模型;血浆中Cmax是关节液Cmax的1.53倍。结论:血液及关节液中均存在青藤碱,但在血液中的Cmax高于关节液。
OBJECTIVE To investigate the pharmacokinetic parameters of sinomenine(SIN)in the blood and joint fluids of rabbits under physiological state.METHODS 100 mg·kg~(-1) of SIN was orally administered to rabbits.Joint fluids were collected by microdialysis,meanwhile blood samples were collected from the ear veins.The LC/MS/MS method was applied to detect the concentrations of SIN in the joint fluids and blood.Pharmacokinetics parameters were calculated by DAS 2.1.1 software.RESULTS The main pharmacokinetics parameters of SIN in blood were Cmax:2867.5±246.76 ng·mL~(-1),T1/2 Ka:0.31±0.10 h,Ka:3.72±2.4 h~(-1),and T1/2:3.27±0.50 h,which were consistent with the one-compartment model with the first-order elimination kinetics.The main pharmacokinetic parameters of SIN in the joint fluid were Cmax:1 874.688±477.19 ng·mL~(-1),Ka:3.6±0.75 h~(-1),and T1/2:3.91±1.48 h,which were also consistent with the one-compartment model with the first-order elimination kinetics.The Cmaxof SIN in blood was 1.53 times as much as that in the joint fluid.CONCLUSION The Cmaxof SIN in blood is superior to that in the joint fluid.
