目的以莪术醇为底物,用黑曲霉(AS3.739)对莪术醇进行生物转化,并对种子培养基进行优化。方法从接菌量、是否加入氮源、种子培养时间以及转种后的投料时间几方面对黑曲霉的种子培养基进行优化,并从菌浓、pH值及转化率几个方面进行考察,得到优化的培养条件。结果优化后的转化条件为:加入1.8g·L-1的酵母膏,接菌量为3%(9),最佳种子培养时间是24h,最佳投料时间是转种后12h。结论采用优化后的种子培养基,黑曲霉(AS3.739)转化莪术醇为3a-羟基莪术醇(30r-hydroxycurcum01)的转化率明显增大。
Objective To optimize the biotransformation conditions of curcumol by Aspergillus niger AS 3. 739. Methods Biotransformation conditions were optimized by altering the inoculum size, nitrogen resource, inoculum time and the time of adding substrate. Mycelia concentration, pH value and transformation ratio were evaluated for the optimization of the biotransformation conditions. Results The optimized cultivating condition:yeast extract paste was 1.8 g-L-1 inoculum size was 3 %, inoculum time was 24 h, and the time of adding substrate was 12 h after inoculation. Conclusions The biotransformation ratio for curcumol into 3cz-hydroxy curcumol by Aspergillus niger AS 3.739 could be greatly increased under the optimized condition.
