目的探讨鹿茸多肽对辐射诱导大鼠腹部脊髓神经细胞凋亡后caspase-3表达的影响。方法将大鼠腹部脊髓神经细胞进行下列分组实验:对照组(A组),辐射(2Gy)组(B组),辐射(2Gy)+NGF(20ng/ml)组(c组),辐射(2Gy)+鹿茸多肽(400mg/ml)组(D组)。应用2Gv辐射诱导大鼠腹部脊髓神经细胞凋亡后,通过RT-PCR和Western印迹观察caspase-3表达情况。结果B组caspase-3表达上调,D组表达量降低,D组和c组均接近A组的表达量。结论鹿茸多肽使辐射诱导大鼠腹部脊髓神经细胞凋亡后caspase-3表达下降,说明鹿茸多肽具有抑制辐射诱导大鼠腹部脊髓神经元凋亡的作用。
Objective To study the express of caspase-3 on apoptosis of rat abdominal nerve cells in the spinal cord induced by radiation which were treated with the velvet antler polypeptides. Methods The rat abdominal nerve ceils in the spinal cord was divided into normal (groupA), radiation (2 Gy) with treatment the velvet antler polypeptides ( 400 mg/ml ) ( groupB ), radiation ( 2 Gy) with treatment NGF(20 ng/ml)(groupC) and radiation groups(2 Gy)(groupD). After 2 Gy radiation induced apoptosis of rat abdominal nerve cells in the spinal cord,the expression of caspase-3 was tested by RT-PCR and Western blot. Results The expression of caspase-3 was increased in groupB,reduced in groupD,the expression of caspase-3 on groupD and groupC was nearing to groupD. Conclusions The velvet antler poly- peptide might reduce the expression of caspase-3 on apoptosis of rat abdominal nerve cells in the spinal cord induced by radiation. The velvet antler polypeptide has the effect of restraining radiation induce rat abdominal nerve cells in the spinal cord to apoptosis.
