为探讨filipin对人脐静脉内皮细胞(HUVECs)中小凹蛋白-1(Cav-1)的亚细胞定位影响,为进一步研究Cav-1作用机制提供理论依据。培养2~4代HUVECs随机分为:对照组;filipin处理组;采用Na_2CO_3蔗糖密度梯度离心法提取HUVECs中含caveolae的膜碎片,Western blot技术鉴定Caveolae和检测Cav-1蛋白表达。结果显示,与Control组比较,filipin处理组caveolae区Cav-1表达减弱(P〈0.05)。由此可知,Cav-1富集于HUVECs的caveolae,filipin可抑制Cav-1在小凹的表达。
To investigate the effect of filipin on the subcellular localization of caveolin-1(Cav-1) in human umbilical vein endothelial cells(HUVECs),which provide a theoretical basis for further study of Cav-1 functional mechanisms.2-4 generations of HUVECs were cultured and randomly divided into control group and filipin group;caveolae~enriched membrane fractions were isolated from HUVECs by Na_2CO_3 sucrose density gradient method;caveolea was identified and Cav-1 protein expression was detected by Western blot.Compared with control group,the Cav-1 protein expression in caveolea was reduced n filipin group(P 0.05).Cav-1 is richly located in caveolae in HUVECS,and filipin reduces the expression of Cav-1 in caveolae.
