膜片钳实验中,玻璃电极本身特性对获得的实验数据的质量有很大的影响。对于我们常用的直径在10-20微米的细胞来说,使用普通两部拉制或者三步拉制法获得的玻璃电极的物理性质足以很好满足实验需要。但是如果实验中所用的细胞小一些,如线虫细胞直径在3-5微米左右,用普通拉制方法获得的玻璃电极几乎无法使用。这里我们使用压力抛光技术得到了物理性质足够好的电极,在线虫细胞上进行了膜片钳实验。
In patch clamp experiment, the character of the glass pipette is critical to the quality of data. The glass pipette we got using the normal "two step" or "three step" method is suitable to the cells with a diameter of 10-20μM. But if the cells are smaller, for example the diameter of the C.elegans cells is 3-5μM, the glass pipette obtained by the ordinary is almost useless. Here we hired the pressure polishing technique to get the excellent pipette and record signals on the C.elegans cells.
