目的研究防己茯苓汤(防己、黄芪、桂枝、茯苓、甘草)的抗炎活性组分及其作用机理。方法利用水煎、乙醇提取和有机溶剂萃取方法获得水/醇提取物和组分,利用脂多糖和IFN-γ刺激造成鼠巨噬细胞炎症模型,采用Griess法考察细胞中一氧化氮产量,FRAP法考察总抗氧化能力;WesternBlot法考察诱导型一氧化氮合酶(iNOS)和环氧合酶2(COX-2)蛋白的表达。结果和其它组分相比,乙醇-乙酸乙酯提取组分(EAFD)抑制亚硝酸盐的活性最强,其IC50最低,且对静息巨噬细胞的活力无影响。该组分能抑制iNOS(P〈0.01)和COX-2(P〈0.05)的蛋白表达,同时提高总抗氧化能力(P〈0.01)。结论实验结果提示乙醇-乙酸乙酯提取组分是防己茯苓汤主要的抗炎活性组分,部分通过抑制炎症中关键诱导型合酶的蛋白表达,提高总抗氧化能力而发挥抗炎作用。
AIM To study the active constituents and anti-inflammatory mechanism of Fangji Fuling Decoction (Stephaniae tetrandrae Radix, Astragali Radix, Cinnamomi Ramulus, Poria, Glycyrrhizae Radix et Rhizoma ). METHODS Water-soluble and ethanolic extract was prepared and constituents extracted by petroleum ether, ethyl acetate, n-butanol, water were separated. Inflammatory cell model was developed by the combination of IFN-γ with LPS stimulating RAW 264. 7 macrophage. Griess reaction was used for nitric oxide accumulation, MTT assay for cell proliferation ability, FRAP assay for total antioxidant capacity, and Western blot for protein expression. RESULTS Compared with other constituents, ethyl acetate fraction from Fangji Fuling Decoction ethanol (EAFD) extract exhibited the strongest suppressing effect on nitric oxide formation in dose-dependent manner and no cytotoxicity in resting macrophage cells. The protein expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were inhibited by EAFD, at the same time increased total antioxidant capacity. CONCLUSION These results suggest that EAFD may provide potential anti-inflammatory effect through inhibiting iNOS and COX-2 protein expression and increase the total antioxidant capacity.
