目的:观察多重病毒感染后病毒性心肌炎小鼠心肌组织中不同趋化因子的表达,以了解多重病毒感染致病毒性心肌炎的可能机制。方法:120只雄性4周龄Balb/C小鼠随机分3组,A组(40只)每只小鼠腹腔注射0.1mL含100 TCID50的CVB3的Eagle's培养液,建立VMC模型;B组(40只)每只小鼠腹腔注射0.1mL舍100 TCID50的cVB3的Eagle培养液及小鼠巨细胞病毒(MCMV)稀释液0.1mL、腺病毒(ADV)稀释液0.1mL,造成多重病毒感染的病毒性心肌炎小鼠模型;C组(40只)每只小鼠腹腔接种0.1mL不舍病毒的Eagle's培养液作为正常对照组。病毒感染1周后,存活小鼠行体表超声心动图检测后眼球取血处死动物,用化学发光法检测cTnI,用RT—PCR方法检测心肌组织中MCP-1、IP10、MCP-5及MIp-18mR—NA表达水平。结果:多重病毒感染性心肌炎小鼠,主动脉血流流速峰值Vp明显减低,其外周血cTnI检测值明显高于单一病毒感染小鼠,两者有显著差异(P〈0.05)。MCP-1 mRNA的表达水平在多重病毒感染的心肌组织中明显高于正常心肌组织及单一病毒感染的心肌组织(P〈0.05),并且与心肌病变程度呈正相关(r=0.64,P〈0.05);与cTnI的浓度也呈正相关(r=0.61,P〈0.05)。结论:多重病毒感染可影响趋化因子的表达水平,尤其是趋化因子MCP-1 mRNA的表达与病变严重程度、心肌细胞的损伤、心功能改变以及疾病的预后密切相关。
Objective:In this study, we examined the expression of chemokines in multiple viruses infected myocardium tissues, and further explore the possible mechanism of chemokine on the multiple viral myocarditis. Methods:Four-week-old male Balb/c mice were inoculated intraperitonelly with 0.1 mL of CVB3 supernatant in MEM Eagle's solution. The other mice group were inoculated with 0.1ml of CVB3 supernatant in MEM Eagle's solution, 0. 1 mL of MCMV supernatant in MEM Eagle's solution and 0. 1 mL of ADV supernatant in MEM Eagle's solution. Left ejection fraction, segmental motion, indexes of pulse Doppler of aortic flow were examined by echoeardiography in each of mice before sacrifice. The level of serum troponin Ⅰ was monitored quantitatively by using chemiluminescence method to show the myocardium damage. RT-PCR was carried out to synthesize and amplify the cDNA of MCP-1, MIP-1/3, IP-10, MCP-5 and GAPDH according to the manufacturers instruction of PCR kit. Results: The cardiac function indexes (Vp) were obviously decreased in multiple viruses infected myocarditis. The level of serum cTnI in peripheral blood of multiple viruses infected mice were significantly higher than that in single virus infected mice. The expression of MCP-1 mRNA in multiple viruses infected myocardium was significantly higher than that in single virus infected myoeardium. The expression of MCP-1 existed a significant positive correlation with myocardial histopathological score (r = 0.64, P〈0.05) and with the level of cTnI (r = 0.61, P〈0.05). Conclusion: Some chemokines mRNA could express in multiple viruses infected myocardium tissues, especially the expression level of MCP-1 which showed significant correlation with the development of myocardial damage.
