目的探讨神经干细胞向神经元定向分化过程中溶酶体和微管蛋白(β—tublin)的相关性。方法采用细胞培养技术、荧光免疫细胞化学技术以及共焦激光扫描显糍镜挂朱对神经干细胞向神经元定向分化过程中溶酶体和微管蛋白的分布变化进行观察。结果在神经干细胞向神经元定向分化的不同时期,存在着溶酶体与微管蛋白的分布变化,CLSM扫描显示两者在分化初期以核周附近分布明显,随神经元的成熟散在分布于胞质中及突起内。结论在神经干细胞向神经元定向分化过程中溶酶体与微管蛋白关系密切,共同参与细胞内物质的运输。
[Objective] To study and discuss the relativity of lysosome and β-tubulin in neuron-oriented differentiation process of neural stem eels. [Methods] Using the techniques of cell calturo, fluorescent immunocytochemistry and CLSM, we observed the distribution changes of lysosome and tubulin in neuron-oriented differentiation process of neural stem ceils. [Results] During the different phases of neuron-oriented differentiation process of neural stem ceils, there were distribution changes of lysosome and tubulin, and CLSM scanning showed that in the early phase of differentiation they both had an obvious distribution around nuclei, then scattered in cytoplasm and processes as the maturation of neurons. [Conclusion] Lysosome is closely related to tubulin in neuron-oriented differentiation process of neural stem ceils, and they both take part in the transportation of intracellular matter.
