目的该文探讨白介素-17A(IL-17A)rs8193036C/T、rs2275913A/G与干扰素调节因子5(IRF-5)rs729302A/C基因多态性与福建泉州地区人群类风湿关节炎(RA)的易感性关联。方法采用病例-对照研究法,自2014年3月—2016年4月收集泉州正骨医院确诊RA患者108例(病例组)和149名健康对照者(对照组),应用三引物全血扩增法(AS-PCR)对IL-17A rs8193036,rs2275913和IRF-5 rs729302进行基因分型,并用SPSS 18.0统计学软件进行数据结果分析。结果 RA组RF-阳性率、CRP-阳性率、ACCPA-阳性率分别为:88.89%、28.70%、72.22%,DAS28为(4.86±1.63)。IL-17A rs8193036C/T位点的3个基因型CC、TT、CT在健康对照组的分布频率分别为57.0%、16.8%、26.2%,在RA组的分布频率分别为61.1%、6.5%、32.4%,RA组的基因型频率与健康对照组相比,差异有统计学意义(P=0.042),其中携带TT纯合子基因型的RA患病风险较高。IL-17A rs2275913 G/A位点的3个基因型AA、GA、GG在健康对照组的分布频率分别为23.5%、46.3%、30.2%,在RA组的分布频率分别为13.0%、52.8%、34.3%,RA组基因型频率与健康对照组相比,差异无统计学意义(P=0.106)。IRF-5 rs729302A/C的AA、AC、CC基因型在健康对照组的分布频率分别为50.3%、39.6%、10.1%,在RA组的分布频率分别为47.7%、43.1%、8.3%,两组间比较差异无统计学意义(P=0.781)。结论IL-17A rs8193036C/T可能与泉州地区人群RA发病有弱相关性,其中IL-17A rs8193036中TT基因型携带者可能增加RA疾病的易感性。而IL-17A rs2275913A/G和IRF-5 rs729302A/C与泉州地区人群RA发病无相关性。
Objective To study the correlation between the interleukin-17A(IL-17A)rs8193036C/T,rs2275913A/G,(IRF-5) rs729302A/C genetic polymorphism and susceptibility of rheumatoid arthritis in Quanzhou region. Methods 108 cases of RA patients and 149 cases of healthy people in our hospital from March 2014 to April 2016 were selected and respectively used as the case group and control group, and AS-PCR was used for genotyping of IL-17 A rs8193036, rs2275913 and IRF-5 rs729302, and the data results were analyzed by the statistical software SPSS 18.0. Results The positive rates of RF, CRP and ACCPA in the RA group were respectively88.89%,28.70%,72.22% and DAS28 was 4.86±1.63, and the distribution frequencies of three gene types of CC,TT,CT in the IL-17 A rs8193036C/T sites in the healthy control group were respectively 57.0%,16.8%,26.2%, and the distribution frequencies in teh RA group were respectivly 61.1%,6.5%,32.4%, and the difference in the gene frequency between the RA group and healthy control group was statistically significnat(P=0.042), and the prevalence risk of RA with TT homozygote genotype was higher and teh distribution frequencies of the three genetypes AA,GA,GG in the IL-17 A rs2275913 G/A sites in the healthy control group were respectively23.5%,46.3%,30.2%, and the distribution frequencies in the RA group were respectively 13.0%,52.8%,34.3%, and the differences between the two groups were not statistically significant(P=0.106), and the distribution frequencies of AA,AC,CC of IRF-5 rs729302A/C in the healthy control group were respectively 50.3%,39.6%,10.1% and in the RA group were respectively 47.7%,43.1%,8.3%, and the differences between the two groups were not statistically significant(P=0.781). Conclusion IL-17 A rs8193036C/T may have a weak correlation with the RA incidence in population in Quanzhou region, and TT genotype carriers of IL-17 A rs8193036 may increase the susceptibility of RA disease, and IL-17 A rs2275913A/G and IRF-5 rs729302A/C have no correlation with the RA inciden
