目的:通过建立小鼠帕金森病模型,观察丙戊酸钠(valproate,VPA)对中脑黑质多巴胺神经元及胶质源性神经营养因子(glial-derived neurotrophic factor,GDNF)表达的影响,以期探索丙戊酸钠的神经保护作用机制。方法:采用免疫组织化学方法检测小鼠中脑黑质多巴胺神经元;原位杂交方法检测纹状体、海马、顶叶皮质GDNF的表达。结果:GDNF在模型组和VPA组表达明显增加。结论:VPA可能通过上调GDNF的表达而起到神经保护作用。
Objective:To investigate the expression of glial-derived neurotrophic factor(GDNF) in mice with Parkinson's disease,and to study the neuroprotective effects of valproate(VPA).Methods:The animals were randomly divided into four groups,including normal saline(NS)+NS group,NS+MPTP group,VPA +MPTP group and VPA+NS group,with 6 mice in each group.Tyrosine hydroxylase(TH) in the substantia nigra pars compacta(SNc) was measured by immunohistochemistry.The expression of GDNF in striatum,hippocampus and cerebral cortex was observed by in situ hybridization.The A values of positive neurons of GDNF was scaned and analyzed statistically.Results:The expression of GDNF was significantly increased in normal saline(NS)+MPTP group,VPA +MPTP group and VPA+NS group compared to NS+NS group.Conclusion:Protective effect of VPA against Parkinson's disease may be associated with increased expression of GDNF.
