目的研究整合素α9β1对小鼠角膜缝线术后新生淋巴管及血管内皮生长因子C(VEGF-C)表达的影响并探讨其意义。方法 80只BALB/c小鼠,随机分为正常对照组、缝线对照组、α9β1组、α9β1抑制组,每组20只。正常对照组不作特殊处理;缝线对照组、α9β1组、α9β1抑制组小鼠制作双眼角膜缝线模型,术后分别予以左氧氟沙星、左氧氟沙星+α9β1、左氧氟沙星+α9β1单抗Y9A2点双眼,2次/d,共14d。术后第3、5、7、14、21天每组分别取2只小鼠,行角膜免疫荧光淋巴管染色、免疫组织化学染色,观察新生淋巴管的动态变化并计数;RT-PCR、Western blotting法检测不同时间点角膜中VEGF-C的表达。结果正常对照组角膜无新生淋巴管;其余3组小鼠缝线后有新生淋巴管从角膜缘发出,淋巴管计数(LVC)在第14天达峰值。缝线对照组术后第3天开始出现新生淋巴管,第14天LVC为3.27±0.25;与缝线对照组比较,在各观察时间点α9β1组LVC均增多,第14天时LVC为4.93±0.38(P〈0.05),α9β1抑制组角膜新生LVC明显降低,第14天时LVC为2.25±0.34(P〈0.05)。RT-PCR和Western blotting检测结果显示,正常对照组有少量VEGF-C表达,缝线对照组VEGF-C表达较正常对照组显著增多且在第14天达峰值(P〈0.05);与缝线对照组比较,α9β1组VEGF-C表达增高(P〈0.05),而α9β1抑制组VEGF-C表达降低(P〈0.05)。结论整合素α9β1可促进角膜新生淋巴管发生,通过Y9A2抑制整合素α9β1可下调VEGF-C的表达并显著减少角膜新生淋巴管。
Objective To research the effect and clinical significance of integrin α9β1 on the corneal lymphangiogenesis and expression of vascular endothelial growth factor C(VEGF-C) after corneal suture.Methods 80 BALB/c mice were divided into 4 groups, namely normal control group, suture control group, α9β1 group, α9β1 suppression group, with 20 mice in each group.Normal control group received no treatment.Corneal suture was performed in the suture control group,α9β1 group,α9β1 suppression group,and they were treated with levofloxacin,levofloxacin+α9β1,levofloxacin + anti-α9β1 monoclonal antibody(Y9A2), twice every day for 14 days,respectively.A random sample was taken from 2 mice in each group on 3rd,5th,7th,14th,21st day after suture.Corneal lymphangiogenesis was examined by immunofluorescence staining and immunohistochemical method.RT-PCR, Western blotting were used to detect the expression of VEGF-C.Results Normal cornea showed no lymphatic vessels,but new lymphatic vessels were found in corneal limbus of three suture groups,and lymphatic vessels count(LVC) reached peak value on the 14th postoperative day.New lymphatic vessels were found in corneal limbus of suture control group on the 3rd postoperative day, and the LVC was 3.27±0.25 on the 14th day. Compared with suture control group, the LVC of α9β1 group (4.93±0.38) was obviously higher on 14th postoperative day(P0.05),and the LVC of α9β1 suppression group(2.25±0.34) was noticeably lower than that in suture control group on 14th postoperative day(P0.05).Little VEGF-C expressed in normal cornea,and the expression of VEGF-C was higher in suture control group than in normal control group,reaching the peak level on 14th day(P0.05).
