目的:研究中药肉苁蓉对神经母细胞瘤细胞烟碱型乙酰胆碱受体表达的影响及其对抗β淀粉样蛋白(AB)神经毒性作用机制,以探讨其在阿尔茨海默病(alzheimer’s disease,AD)治疗中的作用。方法:比色法测定细胞MTT还原率筛选肉苁蓉安全浓度范围,并用Aβ25~35处理细胞后,测定MTT还原率,蛋白质印迹法(Western blotting)测定烟碱型乙酰胆碱受体a3、a7蛋白表达的变化;TBA比色法测定脂质过氧化产物(丙二醛)的水平。结果:肉苁蓉能提高细胞a3及a7烟碱型乙酰胆碱受体亚单位蛋白质表达水平;并能对抗Aβ25~35引起的a3和a7受体亚单位蛋白质水平降低;同时能减弱Aβ25~35引起的细胞损伤和脂质过氧化水平升高。这些作用随浓度的增加而增强。结论:肉苁蓉能上调烟碱型乙酰胆碱受体亚单位蛋白表达水平,且能对抗Aβ25~35明,的神经毒性作用,在AD的治疗中可能起到一定作用。
OBJECTIVE To investigate the effects of Herb Cisanches on the expression of nicotinic receptor on neuroblastoma SH-SY5Y cells and the mechanism of protective effect on neurotoxicity induced by beta-amyloid peptide (Aβ). METHODS The SH-SY5Y cells were tieated by different concentrations of Herb Cistanches prior the exposure of the cells to Aβ25 35. MTT reduction as,;ay was carried out to understand the influences of the drugs on cellular viability; expressions of nicotinic re ceptor subunits (a3 and a7) at protein levels were detected by Western-blotting; lipid peroxidation was measured by TBARS to observe the antioxidant results by the treatment of the drugs, RESULTS Herb Cistanches stimulated an increase of a7 and a3 proteins in the cultured cells; attenuated the decreased expression of a3 and a7 nAChR subunit proteins and the increased lipid peroxidation on SH-SY5Y cells induced by Aβ, CONCLUSION Herb Cistanches may pay neuroproteetive role by the stimula tion of nAChR expression, which might be important in a therapeutic strategy to AD.
