目的:研究环瓜氨酸肽(CCP)辅助完全弗氏佐剂(CFA)造模对佐剂性关节炎模型大鼠临床体征、血清因子、病理的影响。方法:SD大鼠随机分为4组,即空白对照组、完全弗氏佐剂雄性组(雄CFA组)、完全弗氏佐剂雌性组(雌CFA组)、完全弗氏佐剂+环瓜氨酸肽组(CCP+CFA组)。除空白对照组注射0.1ml生理盐水外,雄CFA组和雌CFA组注射0.1ml CFA,CCP+CFA组注射含有100μg环瓜氨酸肽的0.1ml CFA,造模后每隔3d观察大鼠足肿胀度,第28d处死取血清ELISA法检测IL-6、TNF-a、RF、anti-CCP,致炎侧踝关节进行病理检测。结果:与空白对照组比较,雄CFA组、雌CFA组和CCP+CFA组大鼠关节肿胀度在各个时间段均显著增大,但各模型组之间无显著的统计学意义;与空白对照组比较,各模型组IL-6、TNF-a、RF、anti-CCP均明显升高,具有极显著的统计学意义,与雄CFA组比较,CCP+CFA组IL-6、TNF-a、anti-CCP含量均升高,具有显著的统计学意义,但雌、雄CFA组之间无显著差异;与空白组比较,各模型组炎性细胞浸润、滑膜组织增生、纤维组织增生、巨噬细胞增生、骨质侵蚀水平均明显升高,具有显著的统计学意义,但各模型组之间无统计学意义。结论:环瓜氨酸肽作为一种类风湿关节炎造模的辅助剂,其血清IL-6、TNF-a、RF、anti-CCP均较普通模型显著,可以应用到类风湿关节炎的研究中。
To study the clinical signs, serum factors and pathology of adjuvant arthritis rats, which were induced by complete freand- adjuvant (CFA) with cyclic cindlinated peptide (CCP). Method: Sprague-Dawley rats were randomly divided into four groups: control, male complete freund's adjuvant(maleCFA), female complete freund's adjuvant( femaleCFA), complete freund's adjuvant with cyclic cirullinated peptide( CCP + CFA). maleCFA and femaleCFA were induced by 0. lml CFA. CCP + CFA were induced by 0.1 ml CFA with 100μg CCP. Control animals were similarly injected with saline. During the experimental period, paw swelling was regularly measured every 3 days. All animals were sacrificed on the 28th day after injection, the serum IL-6, TNF-a, RF and anti-CCP levels were measured by ELISA kits, and hind preinflammatory ankle for pathological examination. Result: Compared with the control group, all model groups 'paw swelling obviously increased on every periods( P 〈0.001 ), but the models were no significant among them( P 〉 0. 05). The serum IL--6, TNF-a, RF and anti- CCP levels of all model groups increased contrasting with the control group( P 〈0. 001 ). The IL-6, TNF-a, RF and anti-CCP of CCP + CFA group notably increased comparing with maleCFA group( P 〈0.05), but the maleCFA and femaleCFA were not significant between them( P 〉 O. 05). Inflammatory cell infiltration, synovial hyperplasia, fibrosis, macrophage proliferation and bone erosion levels of all model groups obviously increased contrasting with the control group(P 〈 0.01 ), but the models have no significance among them( P 〉 0.05). Conclu- sion: CCP is an adjuvant for rheumatoid arthritis, mixing with CFA which makes significant contributions to the serum IL-6, TNF-a, RF and anti-CCP levels, and can be used for the experimental study of rheumatoid arthritis.
