细胞凋亡失控是肿瘤的一个标志,与多种恶性肿瘤的发生和发展相关,肿瘤细胞内肿瘤凋亡抑制蛋白(IAP)家族的表达上调正是肿瘤细胞产生辐射抗性和耐药性的基础。Smac来源于线粒体,能特异地作用于IAP家族的XIAP,解除其凋亡抑制作用,从而激活半胱氨酸蛋白酶,发挥其促细胞凋亡作用,对肿瘤细胞有辐射和化疗増敏作用,可能成为肿瘤治疗的新方法和新途径,是目前相关领域的研究热点。
Deregulation of apoptosis is a hallmark of cancer and contributes to development and progression of various malignancies. Upregulation of inhibitor of apoptosis protein(IAP) family in tumor cells is the basis of resistance to radiotherapy and chemotherapy. Smae,a mitoehondria protein released during apoptusis to activate easpase by inhibiting cellar inhibitor of apoptosis protein-XIAP of IAP family, so plays a pro-apoptotic role, then in turn results in tumor cells radiosensitization and chemosensitization, which may become a new approach for tumor treatment, and is currently the hotspot of the research field.
