本研究为筛选胆管癌癌变的标志物,开发相应的药物研究,通过对胆管癌血清双向电泳的样品处理、pH范围、上样量的选择、电泳参数的设置和SDS凝胶电泳染色方法等进行比较优化.确定了样品预处理后通过Millpore公司推出的Montage Albumin Depletekit去除血清中高丰度的白蛋白,再经丙酮沉淀,用裂解液溶解后,采用pH3.5-10与pH5-8的两性载体电解质以体积比1:4混合使用,100μg的上样量,12.5%的SDS-PAGE胶,16mA/胶进行双向电泳的电泳体系.优化了胆管癌血清双向凝胶电泳的条件,得到了分辨率较好的胆管癌血清的蛋白图谱,为进一步的研究奠定了基础.
The aim of this study is identify candidate biomarkers for Cholangiocarcinoma in clinical serum samples. Efforts were made to establish the two dimensional gel electrophoresis (2-DE) separation of serum proteins. This efforts were focused on sample preparation, pH range selection ,loading amount, electrophoresis parameters optimization, and protein staining. The results revealed that high quality protein samples were obtained by TCA/acetone precipitation after depletion of high-abundant proteins using Montage Albumin Deplete kit ( Millipore). High resolution, reproduction 2-DE maps were obtained when the ration of Amepline pH 3.5 - 10 to pH 5 - 8 was set at 1:4 with 100 /a,g proteins each, and run under 16 mA. The optimized conditions for serum protein in two-dimensional gel electrophoresis of the Cholangoeareinoma are the foundation for further study on serum differential proteomics.
