以金铁锁红色愈伤组织为材料,用1%盐酸-甲醇提取花色苷,研究其在不同温度、光照、pH、可溶性糖、还原剂、氧化剂、防腐剂和金属离子等条件下的稳定性。实验结果表明,金铁锁花色苷在温度为4~60℃及偏酸性条件下较稳定;光照对花色苷具有较强的破坏作用,应避光存放;氧化剂和抗坏血酸对其影响较大,但在还原剂作用下较稳定。此外可溶性糖及防腐剂对愈伤组织有一定的增色作用;K+、Zn2+、Mn2+、Mg2+、Ca2+的存在对其稳定性没有较大影响,但Cu2+、Fe3+对其有明显的破坏作用,因此在储存愈伤组织或花色苷时应避免使用铜器和铁器。
The anthocyanin was extracted by 1% HCl- ethanol from the red callus of Psammosilene tunicoides. Effects of temperature,light,pH value,soluble sugar,reductant,oxidant,antiseptic,metalions and other conditions anthocyanin stability was studied. The results showed that anthocyanin was stable at temperature 4 ~ 60℃ and acidic condition. Light had strong damage effect for anthocyanin stability,so anthocyanin should be kept away from light. Anthocyanin was greatly affected by the oxidant and Vc,but was well tolerated to reductant. Moreover,the soluble sugar and antiseptic had certain hyperchromic effect for the callus. K+、Zn2+、Mn2+、Mg2+、Ca2+ had no great influence,but Cu2+and Fe3+had obvious damage to anthocyanin stability,so the callus or anthocyanin of P. tunicoides should not be stored in copperware and ironware.
