本文利用从棉花(Gossypium hirsutum L.)纤维中获得的生长素结合蛋白(auxin-binding protein)基因cDNA全长,经生物信息学分析,构建了由棉花纤维特异表达启动子E6驱动的该基因过量表达载体(pBE6:GhABPI (GUS)。通过农杆菌介导法将该基因转化棉花,获得了经PCR检测的阳性胚性愈伤组织。为近一步研究生长素结合蛋白在棉纤维发育过程中的功能奠定了基础。
In order to study the function of auxin binding protein during the development of cotton (Gossypiurn hirsutum L.), the construction of plant-expressed vector (pBE6: : Gh,4BP(GUS )) of Auxin binding protein drived by the fiber-specially expressed promoter E6 which had been cloned from cotton fiber had been finished and introduced into cotton calli through Agrobacterium-mediated transformation. Kanamycine screening and PCR assay initially confirmed that the transformation of GhABP into the transgenic cotton calli. Our work represented a first step towards function analysis of GhABP in the development of cotton fiber.
