探讨昆布多糖对人结肠癌LoVo细胞增殖的影响以及凋亡相关的Caspase-8、Caspase-3、Caspase-7活性的变化.分别以不同质量浓度的昆布多糖处理LoVo细胞,采用Hoechst 33258染色在荧光倒置显微镜下观察细胞形态变化;Caspase-8、Caspase-3、Caspase-7活性检测试剂盒检测Caspase-8、Caspase-3、Caspase-7活性.昆布多糖作用LoVo细胞72 h后,随着昆布多糖质量浓度的增高(400,800,1 600μg/mL),凋亡的细胞数也随之增加.昆布多糖作用LoVo细胞24 h后,随着昆布多糖质量浓度的增高(400,800,1 600μg/mL),Caspase-8、Caspase-3、Caspase-7的活性逐渐升高并存在剂量依赖性(P〈0.05).昆布多糖具有明显的诱导细胞凋亡的作用,且伴随Caspase-8、Caspase-3、Caspase-7活性升高通过Caspase信号传导通路诱导LoVo细胞凋亡.
To study proliferation effect of laminarin on human colon cancer LoVo cell and activity of apoptosis-related Caspase 8,3,7.Deal with LoVo cells under different concentrations of laminarin by Hoechst 33258 staining is observed under fluorescence microscope in morphological changes;Caspase-8,Caspase-3,Caspase-7 activity assay kit Caspase-8,Caspase-3,Caspase-7 activity.Laminarin role in LoVo cells after 72 h,with the laminarin concentration increase(400,800,1 600 μg/mL),apoptotic cells also increase.Laminarin role in LoVo cells after 24 h,with the laminarin concentration increase(400,800,1 600 μg/mL),Caspase-8,Caspase-3,Caspase-7 activity increase gradually and in a dose dependent manner(P0.05).Laminarin significantly induce apoptosis,and with Caspase-8,Caspase-3,Caspase-7 activity increase Caspase signaling pathways induced by LoVo cells.
