目的:探讨中药丹皮酚对人甲状腺未分化癌FRO细胞相关lncRNA表达的影响。方法:选取不同浓度的丹皮酚作用于人甲状腺未分化癌FRO细胞,采用MTT法检测细胞的存活率,对细胞形态学的改变采用倒置相差显微镜观察,qRT-PCR检测不同浓度的中药丹皮酚作用于FRO细胞后H19、HOT、PCA三种癌症相关lncRNA的表达水平。结果:不同浓度的丹皮酚能够在不同程度上抑制甲状腺未分化癌FRO细胞的生长,经过药物处理后48h,检测发现FRO细胞中H19、HOTAIR、PCA3的表达显著增高,且随浓度增加而逐渐升高。结论:丹皮酚可以抑制甲状腺未分化癌细胞的生长,不同浓度的丹皮酚对甲状腺未分化癌细胞中的lncRNA表达有影响,说明中药提取物丹皮酚可能通过lncRNA对肿瘤的发生发展产生影响,相关机制需要进一步研究。
Objective: To investigate whether the traditional Chinese medicine Paeonol can affect the expressive level of lncRNA in human thyroid cancer cell line FRO. Methods: Human thyroid cancer cell line FRO was incubated with different concentrations of paeonol for 48 h. Then cell survival rate was measured by MTT assay and cell morphology change was observed by phase contrast microscope. The expressive level of cancer-related lncRNA( H19,HOT,PCA) of human thyroid cancer cell line FRO was measured by qRT-PCR. Results: Paeonol could inhibit the growth of FRO to different degree. After treated with Paeonol for 24 hours,the expression level of cancer-related lncRNA in FRO was increased. In addition,the lncRNA expression increased gradually with the increase of Paeonol concentration. Conclusion:Paeonol can inhibit the growth of FRO. Different concentrations of Paeonol have different effects on the changes of the expressive levels of lncRNA in FRO. Traditional Chinese medicine Paeonol may influence the progression of human thyroid cancer by changing the expression levels of cancer-related lncRNA. Further investigation on the mechanism is needed.
