向培养了7 d的匍枝筋骨草悬浮细胞培养体系内分别添加0.1,0.2,0.3,0.4,0.5 mg/L的ABA,以研究ABA对匍枝筋骨草细胞防御系统的作用及ABA对蜕皮激素合成的影响.试验结果表明:向培养7 d的悬浮体系中,添加0.1-0.4 mg/L的ABA后,悬浮细胞第4天出现褐化;添加0.5 mg/L的ABA后,第3天出现悬浮细胞褐化现象;添加不同质量浓度的ABA后第4天测其悬浮细胞生长量均与对照差异不显著;添加不同质量浓度ABA后悬浮细胞的SOD,POD和CAT活性在48 h内均显著上升,而PAL活性变化在2-24 h间不显著,后上升;添加不同质量浓度的ABA可使筋骨草悬浮细胞的蜕皮激素含量增高,而添加0.1 mg/L ABA,其蜕皮激素含量增高最为显著.
This study focuses on the Ajuga lobata D. Don suspension cell culture system 'after being cultivated for 7 days. In order to study the effect of ABA to the cell defense and/3-ecdysone synthesis of Ajuga lobata D. Don suspension cell culture system, five kinds of different concentration ABA from 0.1 to 0.5 mg/L were added to this system separately. The results show that the Ajuga lobata D. Don suspension cell became brown 4 days after addi- tion of ABA from 0.1 to 0.4 mg/L. The cell became brown 3 days after attition of ABA with 0.5 mg/L. No matter what kinds of ABA were added, the suspension cell growing in the treated culture system have no significant difference compared with controls when observed on the 4th day. No matter what kinds of ABA were added, the activi- ties of SOD, POD and CAT increase when observed during the 48 hours, while the activities of PAL does not change remarkably during 2 to 24 hours, but increase after 24 hours. No matter what kinds of ABA were added, the synthesis of β-ecdysone increase, but the change increases most significantly when 0.1 mg/L ABA was added.
