血管内皮祖细胞参与肺癌组织血管形成的实验研究

目的利用血管内皮祖细胞(endothelial progenitor cells,EPCs)在血管形成过程中的积极作用,研究其参与肺癌组织的血管形成的特点。方法采用外周血单个核细胞体外经定向分化、培养为EPCs,并用4,6-联脒-2-苯基吲哚(DAPI)荧光标记,将DAPI荧光标记的EPCs与人肺癌细胞株SPC-A1同时给BALB/C裸鼠(B组)皮下注射,观测肿瘤生长情况,并与单纯接种SPC-A1裸鼠(A组)比较。结果 EPCs能从人外周血获得,可体外扩增约100倍。接种荷瘤后,A组裸鼠平均寿命为85±9d,B组为78±8d;A组肿瘤平均体积为1.44±0.45cm3,B组为1.83±0.32cm3。A组与B组相比,裸鼠平均寿命和肿瘤体积大小差异均有统计学意义(P〈0.05,P〈0.01)。结论 EPCs能参与肺癌组织血管形成,促进肺癌生长,使裸鼠寿命缩短。

英文摘要:

Objective To make use of endothelial progenitor cell's positive role on the process of angiogenesis,we study the angiogenesis of lung cancer. Methods Endothelial progenitor cells were isolated from PBMC and then in vitro differentiate,culture and amplify for endothelial progenitor cells;To lable endothelial progenitor cells with DAPI fluorescence;To transplant both marked cells and lung carcinoma SPC-A1 cells in BALB/C mice meanwhile to observe endothelial progenitor cells'effect on angiogenesis during tumor growth. Results Endothelial progenitor cells can be obtained by culturing the peripheral blood and amplificated by about 100 times in vitro. Through subcutaneous inoculation, subcutaneous inoculation xenograft tumor mode of SPCA-1 was gotten. Nude mice mean life in group A were 85.±9 days,in group B 78±8days. Gross tumor volume in group A were 1.44±0.45 cm3,in group B 1.83± 0.32 cm3. In the period of observation, the mean life span difference of three groups nude mice had sence(P0.05). The gross tumor volume's difference between group A and group B had statistical significance (P0.01). Conclusions It is suggested that endothelial progenitor cells can participate in lung cancer tube angiogenesis,hasten the growth of lung cancer and shorten the life of nude mice.
