为建立双向电泳方法研究鸡羽髓蛋白组学,以进一步了解病毒与宿主相互作用的新机制,从SPF鸡羽根挤出羽髓,提取其中蛋白进行双向电泳,并对不同裂解液、IPG胶条pH值范围、一向等电聚焦条件、二向SDS-PAGE凝胶浓度等影响因素进行优化。结果显示,采用17cm,pH5~8IPG胶条,400μg羽髓蛋白进行的双向电泳,图谱用PDQuest8.0.1分析,可分辨的蛋白点约为700左右,不同样本间的匹配率大于80%。可见,本研究建立的双向电泳方法分辨率及重复性均较高。
Total proteins were extracted from the feather pulps of SPF chickens and separated under 2-dimentional electrophoresis(DE) to investigate interaction between viruses and hosts.The 2-DE method was established by optimizing different lysis buffers,pH range of IPG,IEF,concentration of polyacrylamide gel etc.About 700 resoluble polypeptide spots could be detected on the map when 400μg proteins of feather pulp were loaded onto 17 cm,pH5-8 IPG strip for 2-DE followed by staining with Coomassie brilliant blue G-250.The match rate of feather pulp maps from individual chickens was higher than 80%.Results demonstrated good reproducibility and resolution of 2-DE mapping of chicken feather pulp proteins.
