木鳖子提取液抗氧化活性的分析

目的用高效液相色谱法(high performance liquid chromatography,HPLC法)测定木鳖子提取液的抗氧化活性。方法苯甲酸与Co2+催化过氧化氢产生的羟基自由基反应,生成多种羟基化物,HPLC法测定加入木鳖子提取液前后苯甲酸改变量,从而间接测定木鳖子提取液对羟基自由基的抗氧化活性。色谱柱:ODS-100 C18(5μm,150 mm×4.6 mm);流动相:甲醇-乙酸铵水溶液(0.02 mol/L,5∶95);检测波长:230 nm;柱温:25℃;流速:1.0mL/min。结果苯甲酸的线性范围为0.08~1.6μg(r=0.999 7),RSD〈0.96%(n=6)。结论木鳖子提取液有一定的抗氧化活性。

英文摘要:

Objective To analyse the antioxidant activity of the extract of Momordia cochinchinensis by high pressure liquid chromatography(HPLC) method. Methods Co^2+ and H2O2 were catalyzed by benizoic to generate the hydroxyl radical reaction which also produced several hydroxylated products.The variable quantity of benizoic acid before and after adding the extract of Momordia cochinchinensis in solution was monitered by using HPLC method,and the antioxidant activity of the extract was determined indirectly.The HPLC analysis was performed on a C18 analytical column maintained at 25 ℃.The mobile phase consisted of 0.02 mol/L ammonium acetate-methanol(95∶5,V/V) delivered at a flow rate of 1.0 mL/min.The UV detection wavelength was set at 230 nm. Results The linear range of benizoic acid was 0.08-1.6 μg(r=0.999 7) with RSD less than 0.96%(n=6). Conclusions The extract of the Momordia cochinchinensis has antioxidant activity.
