落新妇甙对小鼠移植心肌细胞凋亡的抑制作用

目的观察落新妇甙(Astilbin)对小鼠心脏移植急性排除反应中心肌细胞凋亡的抑制作用。方法采用小鼠颈部心脏移植模型。共分3组:对照组供、受体均为C57小鼠;移植组供体为BALB/c小鼠,受体为C57小鼠;落新妇甙组在移植组基础上,心脏移植后每天以落新妇甙1mg/kg灌胃。分别于术后第1、3、5、7天各取4只移植心脏.原位末端标记(TUNEL)法染色检测心肌细胞凋亡,以心肌细胞凋亡阳性细胞数占总心肌细胞数的百分比作为心肌细胞凋亡指数(apoptosis index,AI)。结果移植组心肌细胞于术后第1天即已出现凋亡,第3天明显增加,第7天达到高峰。落新妇甙组各时间点心肌细胞凋亡指数明显小于对应的移植组(P〈0.01)。结论落新妇甙对心脏移植排斥反应中心肌细胞凋亡有显著抑制作用。

英文摘要:

Objective To investigate the inhibitory effect on apoptosis of myocardial cells in mice heart transplantation rejection. Methods The model of mice cervical heterotopic heart transplantation was set up. The mice were randomly divided into three groups. In control group, the donors and accepors were all C57 mice; in transplant group, the donors were BALB/c mice and the accepors were all C57 mice; in astilbin group, mice were treated with same agent as those in transplant group were, except that they were fed with additional astilbin after heart transplantation. On the lst,3rd,Sth,and 7th day after transplantation, apoptosis index was evaluated by the percentage of myocardial cells with TUNEL positive staining. Results The apoptotic myocardial cells appeared on the 1st day and significantly increased at the 3rd day, while the the apoptotic peak was reached on the 7th day after transplantation in transplant group. Apoptosis indices of myocardial cells in astilbin group were significantly lower than those in transplant group (P〈 0.01). Conclusion Astilbin could inhibit the apoptosis of myocardial cells in mice heart transplantation rejection.
