目的探索缺血性脑损伤后,尼莫地平(Nimodipine,NDP)对大鼠神经再塑过程中神经细胞黏附分子(neural cell adhesion molecule,NCAM)表达的影响。方法Wistar大鼠100只,随机分成假手术对照组(sham operation control group,SOCG,n=10)、脑缺血对照组(cerebral ischemia control group,CICG,1、3、7d,n=10),脑缺血高、低剂量尼莫地平治疗组(nimodipine treatment group,H-NTG,L-NTG,1、3、7d,n=10)。手术阻断CICG和NTG右侧大脑中动脉,制造缺血性脑损伤模型,NTG腹腔注射NDP(0.5mg·kg^-1和1mg·k^-1)。分别于1、3和7d脑血流再灌注,神经功能缺损评分、局部脑血流量(regional cerebral blood flow,RCBF)测定后取材,观察大脑表面血管形态,检测NCAM的表达。结果H-NTG3d和7d的PO2高于CICG(P〈0.05或P〈0.01);L-NTG和H-NTG的神经功能缺损评分低于CICG(P〈0.05或P〈0.01),其RCBF的灌注单位和NCAM的表达也高于CICG(P〈0.05或P〈0.01)。结论尼莫地平能改善缺血脑损伤大鼠的神经功能,增加局部脑血流量,缩小缺血脑组织梗死面积,促进NCAM的表达。
Aim To explore the effects of Nimodipine (NDP) treatment on the expression of neural cell adhesion molecule (NCAM) after the ischemic brain injury in rats. Methods One hundred male Wistar rats (180 - 220g) were randomly divided into four groups: sham operation control group ( SOCG, n = 10) , cerebral ischemia control group ( CICG, 1,3,7 d ,n = 30), high and low dose Nimodipine treatment group (H-NTG, L- NTG,1,3,7 d,n = 10). Their middle cerebral arteries of CICG and NTG were blocked by filament forming brain injured, the NTG groups were treated with NDP (0. 5 mg · kg^-1 and l mg · kg^-1), respectively, and then, their cerebral blood flow was reperfused. Their blood vessel shape of cerebri surface was observed and the expression of NCAM was determined, after neurologic impairment(NIP) was scored and regional cerebral blood flow (RCBF) was surveyed, respectively. Results 3 d , 7 d PO2 of H-NTG were higher than those of CICG ( P 〈 0. 05 or P 〈 0. 01 ) ; NIP scores of the L-NTG and H-NTG were lower, RCBF and the expression of NCAM were higher than those of the CICG (P 〈 0. 05 or P 〈 0. 01 ). Conclusions NDP could improve neural function of ischemic brain injury in rats,increase the RCBF, contract the infarct sizes of ischemic brain tissue, and facilitate the expression of the NCAM.
